چکیده

Objective: On account of several complications and adverse effects associated with the use of conventional chemotherapeutic regimen, the advanced drug-targeted therapies have gained the remarkable attention of the researchers due to their fabulous pharmaceutical and therapeutic advantages. The present study was designed with the aim to synthesize manganese ferrite nanoparticles (MnFe2O4 NPs) and folic acid-conjugated doxorubicin (DOX)-loaded manganese ferrite bovine serum albumin NPs (FA-BSA-DOX-MnFe2O4 NPs) using desolvation cross-linking method. Methods: Having assessed their physicochemical characteristics, the prepared NPs were evaluated for hem compatibility, photo-mediated cytotoxicity, and anti-cancer potential against human cervical carcinoma cell line (HeLa) using a range of in vitro assays which include hemolysis assay, sulforhodamine B (SRB) and MTT assays. Results: Spectroscopic characterization revealed that MnFe2O4 NPs were spherical with an average size diameter of approx. 15 nm and a band gap of 1.4 eV. Another remarkable feature of FA-BSA-DOX conjugated MnFe2O4 NPs was high entrapment efficiency (approx. 95%). MTT assay demonstrated that MnFe2O4 NPs revealed potential photosensitizing ability upon exposure to sunlight. FA-BSA-DOX conjugated MnFe2O4 NPs showed promising cytotoxicity against human cervical epithelial malignant carcinoma cell line (HeLa). Interestingly, the cytotoxicity of these NPs was gradually increased with time of exposure to sunlight. Conclusion: These findings suggested that FA-BSA-DOX conjugated MnFe2O4 NPs exhibit promising photosensitizing and anticancer potential against human cervical carcinoma and thus can be considered as an alternative targeted therapy against human cervical cancer.

Folate Receptor-targeted Bioflavonoid Genistein-loaded Chitosan Nanoparticles for Enhanced Anticancer Effect in Cervical Cancers

In this study, novel folic acid-conjugated chitosan nanoparticle was formulated for specific delivery of bioflavonoid, Genistein (GEN), to the cervical cancer cells. The prepared GEN-loaded chitosan nanoparticles (GCN) and folic acid-conjugated GCN (FGCN) showed smaller size with a controlled drug release profile. FGCN exhibited enhanced internalization potential in HeLa cells than that of GCN....

Investigation of cytotoxicity properties of zinc oxide nanoparticles in spherical and rod shaped on leukemia cells

In this study, we reported a method to associate doxorubicin drug on folic acid functionalized SiO2/ZnO nanoparticles (NPs) in rod and spherical shapes. The clinical usage of the anticancer drug, doxorubicin (DOX), is limited by severe side effects and cell resistance. Targeted drug delivery by binding DOX to nanoparticles could overcome these limitations. The surface functionalization of the Z...

Synergistically enhanced selective intracellular uptake of anticancer drug carrier comprising folic acid-conjugated hydrogels containing magnetite nanoparticles

Targeted drug delivery has long been extensively researched since drug delivery and release at the diseased site with minimum dosage realizes the effective therapy without adverse side effects. In this work, to achieve enhanced intracellular uptake of anticancer drug carriers for efficient chemo-therapy, we have designed targeted multifunctional anticancer drug carrier hydrogels. Temperature-re...

Folic-acid-conjugated pullulan/poly(DL-lactide-co-glycolide) graft copolymer nanoparticles for folate-receptor-mediated drug delivery

BACKGROUND Nanoparticles have been extensively investigated for targeted delivery of anticancer drugs. Since the folate receptor is universally over-expressed on the tumor cell membrane, folic acid is often used to modify the fate of nanoparticles in biologicals. METHODS To fabricate targetable nanoparticles, folic acid was conjugated to a pullulan backbone and poly(DL-lactide-co-glycolide) (...
